A novel "Integrated Biomarker Response" calculation based on reference deviation concept ArchiMer
Sanchez, Wilfried; Burgeot, Thierry; Porcher, Jean-marc.
Multi-biomarker approaches are used to assess ecosystem health and identify impacts of environmental stress on organisms. However, exploration of large datasets by environmental managers represents a major challenge for regulatory application of this tool. Several integrative tools were developed to summarize biomarker responses. The aim of the present paper is to update calculation of the "Integrated Biological Response" (IBR) described by Beliaeff and Burgeot (Environ Toxicol Chem 21:1316-1322, 2002) to avoid weaknesses of this integrative tool. In the present paper, a novel index named "Integrated Biological Responses version 2" based on the reference deviation concept is presented. It allows a clear discrimination of sampling sites as for the IBR, but...

A pollution-monitoring pilot study involving contaminant and biomarker measurements in the Seine Estuary, France, using zebra mussels (Dreissena polymorpha) ArchiMer
Minier, Christophe; Abarnou, Alain; Jaouen Madoulet, Agnes; Le Guellec, Anne-marie; Tutundjian, Renaud; Bocquene, Gilles; Leboulenger, François.
Zebra mussel (Dreissena polymorpha) is an invasive species that has proliferated in European and North American rivers and lakes during the last century. In this study, D. polymorpha has been used to provide information on contamination levels and biological effects in the Seine Estuary (France). The bivalves accumulated polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs) to a high degree with values reaching 800 ng/g dry weight for PCBs (sum of 20 congeners), and 1,000 ng/g dry weight of PAHs (sum of 14 compounds) in the whole body. These values are among the highest reported of PCBs and, to a lesser extent, of PAHs in other contaminated areas in the world. Toxic equivalent quantities of PCBs and PAHs detected in zebra mussels...

Active biomonitoring in Greek coastal waters: Application of the integrated biomarker response index in relation to contaminant levels in caged mussels ArchiMer
Tsangaris, Catherine; Hatzianestis, Ioannis; Catsiki, Vassiliki-angelique; Kormas, Konstantinos Ar; Strogyloudi, Evangelia; Neofitou, Christos; Andral, Bruno; Galgani, Francois.
An integrated approach using biomarkers and contaminant levels in mussels Mytilus galloprovincialis L was employed to assess chemical contamination in Greek coastal waters within the framework of the MYTIMED program. Biomarkers (metallothioneins, glutathione S-transferase, catalase, acetylcholinesterase and RNA:DNA ratio) have been previously described in mussels caged at 14 sites in coastal areas influenced by different types of anthropogenic activities. This study applied a biomarker index, the Integrated Biomarker Response (IBR) to summarize biomarker responses and relate stress levels to concentrations of organochlorine compounds (PCBs, DDTs), polycyclic aromatic hydrocarbons (PAHs), and metals (Cu, Ni, Fe, Zn) measured in the mussel tissues. The IBR...

CHAVES,Valesca de Souza; MARCON,Jaydione Luiz; DUNCAN,Wallice Paxiúba; ALVES-GOMES,José Antônio.
ABSTRACT Deltamethrin is a pyrethroid insecticide widely used in pest control in Brazilian agriculture. The intensive and disordered use of this pesticide in the Amazon region can drive it into aquatic ecosystems in several ways, but mainly by runoff and leaching. The present study was conceived to determine the acute toxicity (LC50) of a deltamethrin based pesticide (DBP) and to characterize its effects on two biochemical biomarkers, glutathione-S-transferase (GST) and acetylcholinesterase (AChE), in tissues of the electric fish Microsternarchus cf. bilineatus. Fishes were exposed to concentrations of 1, 2, 3, 4 and 5 μg L-1 of DBP for up to 96 hours. For each treatment, the absolute activity of the enzymes GST (muscle and liver) and AChE (muscle and...

Exposure to aflatoxins (AFs) in the diet may favour the development of hepatocellular carcinoma (HCC) and the acute exacerbation of hepatitis in chronic hepatitis B virus (HBV) carriers. Measurement of biomarkers such as aflatoxin M1 (AFM1), a metabolite of aflatoxin B1 (AFB1), in urine allows for the assessment of populations exposed to aflatoxins. The aim of this study was to investigate the occurrence of aflatoxin M1 in the urine of HBV carrier and non-carrier patients. One group included 43 randomly selected HBV carriers treated at two hospitals in the city of Maringa, Brazil, from March to June 2008. Control group consisted of 29 healthy adult volunteers with anti-HBs positive and HBsAg negative test results. Detection of AFM1 was performed by...

The objective of this study was to develop an integrative approach in ecotoxicology (from biomarkers to population genetics) to assess the ecological status of fish populations. Flounders (Platichthys flesus) collected after the spawning season in the heavily polluted Seine estuary were compared with the moderately polluted Bay of Douarnenez. The muscle energetic reserves were highly depleted in Seine vs. Douarnenez fish. The Seine fish displaying a reduced capacity to manage the oxidative stress and a higher energetic metabolism. An increase in the content of muscle membrane phospholipids (sphingomyelin, phosphatidylserine, free sterols) was detected in the Seine vs. Douarnenez fish. The data integration allowed to hypothesize relationships between...

Are shallow-water shrimps proxies for hydrothermal-vent shrimps to assess the impact of deep-sea mining? ArchiMer
Mestre, N.c.; Auguste, M.; De Sá, L.c.; Fonseca, T.g.; Cardoso, C.; Brown, A.; Barthelemy, D.; Charlemagne, N.; Hauton, C.; Machon, J.; Ravaux, J.; Shillito, B.; Thatje, S.; Bebianno, M.j..
Polymetallic seafloor massive sulphide deposits are potential targets for deep-sea mining, but high concentrations of metals (including copper - Cu) may be released during exploitation activities, potentially inducing harmful impact. To determine whether shallow-water shrimp are suitable ecotoxicological proxies for deep-sea hydrothermal vent shrimp the effects of waterborne Cu exposure (3 and 10 days at 0.4 and 4 μM concentrations) in Palaemon elegans, Palaemon serratus, and Palaemon varians were compared with Mirocaris fortunata. Accumulation of Cu and a set of biomarkers were analysed. Results show different responses among congeneric species indicating that it is not appropriate to use shallow-water shrimps as ecotoxicological proxies for deep-water...

Assessing chronic fish health: An application to a case of an acute exposure to chemically treated crude oil ArchiMer
Mauduit, Florian; Domenici, P.; Farrell, A. P.; Lacroix, C.; Le Floch, S.; Lemaire, P.; Nicolas-kopec, A.; Whittington, M.; Zambonino-infante, Jose-luis; Claireaux, Guy.
Human alteration of marine ecosystems is substantial and growing. Yet, no adequate methodology exists that provides reliable predictions of how environmental degradation will affect these ecosystems at a relevant level of biological organization. The primary objective of this study was to develop a methodology to evaluate a fish’s capacity to face a well-established environmental challenge, an exposure to chemically dispersed oil, and characterize the long-term consequences. Therefore, we applied high-throughput, non-lethal challenge tests to assess hypoxia tolerance, temperature susceptibility and maximal swimming speed as proxies for a fish’s functional integrity. These whole animal challenge tests were implemented before (1 month) and after (1 month)...

Association of genotypes with viral load and biochemical markers in HCV-infected Sindhi patients BJM
Riaz,Saba; Bashir,Muhammad Faisal; Haider,Saleem; Rahid,Naeem.
Abstract The presented study had two objectives. The first was to examine distributions of Hepatitis C Virus (HCV) genotypes in Sindh, Pakistan, where HCV is prevalent. The other was to explore clinically relevant relationships between the genotypes, viral load (measured by real-time polymerase chain reaction assays) and biochemical markers. For this, 1471 HCV-infected patients in six cities in Sindh were recruited and sampled. HCV genotype distributions varied among the cities, but genotype 3a was most prevalent, followed by 3b, 1a and 1b (detected in 51.5, 22.7. 9.25 and 3.2% of the cases, respectively). No type-specific sequences were detected in serum samples from 189 (12.8%) of the 1471 patients. Frequencies of low (<200,000 IU/mL serum),...

Engineered nano-sized Cu oxide particles are extensively used in diverse applications. Because aquatic environments are the ultimate "sink" for all contaminants, it is expected that nanoparticles (NP) will follow the same fate. In this study, two marine invertebrates Scrobicularia plana and Hediste diversicolor were chosen as ecotoxicological models. The aim was to evaluate behavioural (burrowing kinetics, feeding rate) and biochemical (biomarkers) responses of S. plana and H. diversicolor exposed in the laboratory to Cu (10 mu g L(-1)) added in natural seawater either in the form of engineered nanoparticles (NPs) of CuO or as dissolved Cu in 2% HNO(3). Exposure was characterized by considering (i) the physico-chemical fate of NP (ii) the fraction of...

Biological effects of the "Erika" oil spill on the common mussel (Mytilus edulis) ArchiMer
Bocquene, Gilles; Chantereau, Sebastien; Clerendeau, Christelle; Beausir, Emilie; Menard, Dominique; Raffin, Bernard; Minier, Christophe; Burgeot, Thierry; Leszkowicz, Annie Pfohl; Narbonne, Jean-françois.
A 3-year survey was made of several biological markers in mussels (Mytilus edulis) exposed in situ to the oil that came ashore after the wreck of the "Erika" tanker on the Brittany (France) coast in December 1999. The mussel response was assessed using a set of 7 biomarkers, most of them related to the metabolism of organic contaminants. After a series of validation tests, data was evaluated for only 5 biomarkers: acety1cholinesterase (AChE), glutathione S-transferase (GST), catalase (CAT), malondialdehyde (MDA) and deoxyribonucleic acid (DNA) adducts. No significant reductions in GST or CAT levels were observed, levels of DNA adducts and MDA were high during the 6 months immediately following the accident and levels of AChE were significantly lower during...

Biomarker evidence of relatively stable community structure in the northern South China Sea during the last glacial and Holocene ArchiMer
He, Juan; Zhao, Meixun; Li, Li; Wang, Hui; Wang, Pinxian.
High-resolution molecular abundance records for several marine biomarkers during the last glacial and Holocene have been generated for core MD05-2904 (19 degrees 27.32'N, 116 degrees 15.15'E, 2066 in water depth) from the northern South China Sea. The U-37(K') SST record indicates a 4.4 degrees C cooling during the Last Glacial Maximum for this site, consistent with previous reconstructions. The contents Of C-37 alkenones, dinosterol, brassicasterol, and C-30 alkyl diols are used as productivity proxies for haptophytes, dinoflagellates, diatoms, and eustigmatophytes, respectively. These records reveal that both individual phytoplankton group and total productivity increased by several factors during the LGM compared with those for the Holocene, in response...

Biomarkers as tools for monitoring within the Water Framework Directive context: concept, opinions and advancement of expertise ArchiMer

The Water Framework Directive (WFD) currently supports chemical and ecological monitoring programmes in order to achieve the good water surface status. Although chemical and ecological assessments are necessary, they have some limitations. Chemical approaches focus on certain substances identified as priorities, but they do not take into account other potentially harmful substances and also ignore the hazards related to contaminant cocktails. On the other hand, while ecological approaches provide holistic information on the impairment of biological communities in ecosystems, they do not distinguish the role of contaminants in these alterations, and consequently do not allow the establishment of contaminant impact reduction plans. Consequently,...

BACKGROUND: The mortality of lung cancer (LC), increases each year in the world, in spite of any advances, in development of new drugs to advance stages of LC. The high incidence of LC has been associated with smoking habit, genetic diversity and environmental pollution. Antofagasta region has been reported to have the highest LC mortality rate in Chile and its inhabitants were exposed to arsenic in their drinking water in concentrations as high as 870 μg/L. Non-invasive techniques such as biomarkers (Automatic Quantitative Cytometry: AQC and DR70) and Auto Fluorescence Bronchoscopy (AFB) might be potentially useful as a supplementary diagnostic approach and early detection. Early detection is one of the most important factors to intervene and...
